It looks like it should be possible using a program called WiiScrubber. Just google "monster hunter tri custom quests" and look at the first youtube video result, it's got some good instructions.

Black screens usually mean the ISO file is corrupted and therefore unplayable. Something was either wrong with the original ISO you had, or (probably more likely) something got screwed up when you used WiiScrubber. Redump your game to get another ISO file, then try using WiiScrubber again, making sure you follow any steps exactly.

1- extract the quest_03
2-change the mision "team vs a big jaggy" i change the boos with a big baggy
3- then i use wiiscruber again and i replace the quest_03 file with my custom one
4- start the game and it doesnt work .S

Be patient. Answers don't always come when we want them to. It sounds like you're doing something incorrectly when creating a custom quest. Is that how you're normally supposed to make a custom quest? Even if you're just changing Boos to Big Baggy, are you sure there are no other things that need to be changed or other steps that need to be taken? You might also want to ask around the MH3 community for specific answers, not sure anyone here has tried custom quests on Dolphin at least.
