Maybe a strange question, but where did you get the 500GB pen drive?
If it is from Alibaba or the likes you could've been scammed... they sell 500GB pendrives that actually only have 4GB or so because they changed some info. This will allow you to copy up to 500GB of data on the drive but it won't actually be there.
I'm just mentioning this because 500GB pen drives aren't really that common (I've never even seen one in a store and I've lived in multiple countries)

I´ve got from a huge ecommerce site here in Brasil called Mercado Livre (such like an brazilian ebay)
Here is the link:
https://produto.mercadolivre.com.br/MLB-1215606673-pen-drive-512gb-30-pronta-entrega-_JM?quantity=1&variation=35601433089
Besides, I´ve tried with a SD card and the issue continues.
My temporary solution is to get all my ISOs that are on the pendrive, move to my HD and play

512 GB flash drives are pretty common at this point.
They're not the best performance wise, but they're more than enough for things like playing game dumps in Dolphin from. Dolphin only needs to emulate a very slow DVD drive.

I do not know what else to do. I tested it on a Kingston 32GB flash drive, a 1TB Seagate external hard drive, and a kingston 64GB SD, and they all make the same mistake.
Remembering that all ISOs are good, because I open them normally on the PC.
It seems that Dolphin somehow recognizes up to an X number of gigabytes of external drives, because after putting 4 games (on average), this error appears.

Is this on 5.0 stable, or the latest dev build of Dolphin?

Does this also occur on the development version? 5.0 stable is currently almost 3 years old and has had many, MANY changes since then.
Another thing: If you copy the not working ISO/WAD from the pendrive back to an internal harddrive/SSD they start working again?

Tested with the latest version (four hours ago) and yes, all the ISOs working fine after copy/paste.
