I have a Samsung galaxy s6 stock software.
I was wondering if there's any benefit to rooting as far as running dolphin goes. I've been considering cyamogenmod as I approach my warranty expiration

Cyanogen Mod and/or rooting will not make dolphin run better.  
If you actually make use of the root access,maybe you can use that threaded command in ADB if the processors support it,but you can also overclock the CPU and GPU a little after installing something custom with support for it,but keep in mind the battery drain being much faster.
Don't clock it too high or you will fry the device,but other than that it could overheat if set a bit too high.

The CPU core clocks are the most important ones to OC while the GPU can always be a little higher to help.
And if your device is notoriously underclocked below its full potential standard speeds (like Shield TV is with wimpy GPU clock of 200/300Mhz and lower CPU clock),you can set it to those standard rates to solidify your performance,but shorten your battery life due to higher energy consumption.

Edit: Looking at some specs and details,maybe it won't help as much to overclock it seeing how high it already runs at,so only the threaded ADB command should hopefully help a good bit.

I'm still on my lazy azz and here is where I eventually found it mentioned with the root part added to it for functionality.
Problem is,that it is only for Tegra,sorry about mentioning it,but maybe there is an equivalent for Exynos.

Just have to a hope a developer with an Exynos device like yours can find the equivalent command if there is one.
Overclocking in theory is a good idea,  but in practice thermal throttling and general TDP limitations will prevent you from achieving noticeably higher performance in apps like dolphin.
The Mali video driver is always threaded. There is no option to turn it off or on.
I'm the past when using over clocking to play emulators I found it more useful to make barely playable games smoother rather than expecting it to make unplayable ones play.

I've managed to get Luigi's Mansion playing but it's really slow. I'm not rooted at the moment but when I had cyamogenmod on my previous device there were a lot of options for CPU optimisation. Hence the question. I had completely forgotten about straight up over clocking

This is only a NVidia command. To be honest, I have no clue why it's not enabled by default, just like on the desktop.

@OP: Rooting has no effect, but CM may have newer/faster GPU drivers. If so, this will likely help a bit.
