RealXFB is an accuracy option. It was buggy in 4.0; so you may need to use a development build.
You'll also get much closer to the original graphics by using a development version, as 4.0.2 cannot emulate many of the GC's graphical effects correctly, such as distortions. Those changes were made after 4.0.2.

I downloaded the latest dev version and now I don't get a difference between Virtual and Real on the EFB. Is it supposed to be like that or is it because this version overwrote the older version and maybe there's a configuration/ini conflict?
If you are using 1x Native you most likely won't see a difference.
