That is a glovepie script for the classic controller, built for keyboard controls of a specific emulator (yuck!). A better approach is to use a general script that assigns buttons from the classic controller to DInput buttons via ppjoy, thus allowing compatibility with a wide selection of emulators, including Desmume, and use of the analog joysticks. Or better yet, just use a DolphinBar and avoid the hassle of glovepie and ppjoy entirely.

So I can use a Classic Controller with DeSmuME.

Well, the DolphinBar is waaay better, but if you are stuck with glovepie, get a standard ppjoy based script and set it up so you can use it in any emulator. But that's up to you I guess.
All you have to do to create a PIE script is copy paste the code into a text document, and save it as .pie. Open that in glovepie, and there you go.

Purchase DolphinBar, plug it in, and sync the wiimote to it. Set the DolphinBar to Mode3, plug a classic controller into a wiimote, and press a button on the Wiimote. Tada, you have a PC DInput device connected and ready to go. Configure emulator controls, and from that point on all you have to do is just press a button on the wiimote and go. How's that for simple?

If you got a Dolphinbar, you wouldn't have to use it at all. It's a glovepie script, for glovepie only.

Also is DeSmuME X432R based on DeSmuME 9.10?

The first version was based on SVN R5000. Each version is based on the latest SVN the author had available. You can see the history.txt on the Desmume X432R website (no Japanese reading comprehension needed, just look for "r5xxx").
