In the official versions of Dolphin, only if you have a real Wiimote + Motion Plus Accessory or a real Wii Remote Plus (and the other requirements, such as a BT adapter/sensor bar or just the MayFlash DolphinBar). And to be clear, you're free to use unsupported builds (like the ancient ones with rudimentary Motion Plus emulation), it's just you'll need to deal with any problems you encounter by yourself because we don't support unofficial and such old builds here in our forums.
Also, by personal experience, Skyward Sword is a game that makes extensively use of the Motion Plus/Nunchuk/Sensor Bar, while you may actually manage to get outside of the village or even to the first dungeon after messing with emulated Motion Plus controller options for a while, later in the game you'll need to do some complex and quick movements that are virtually impossible to do with the unofficial emulated Motion Plus builds...
