Many game modders have been able to dump and create BRSTMs using various ISO/BRSTM tools such as WiiScrubber, BrawlBox, BRSTM Converter, etc. I'd especially like to load custom audio without the need of an ISO/WBFS extracting tool, which may cause problems if done entirely wrong. However, that may change if a new feature which allows audio to be dumped and loaded into the game becomes implemented to Dolphin, similar to custom textures. It may seem difficult, considering that there are many audio file formats, and the one that we're looking for should be best compatible with Dolphin (and maybe even disk space friendly, too!) Will this be possible? Thanks!
(Feature suggestion) The ability to dump and load custom game audio?

This would be very tricky to do in a generic way, and I don't expect it to become a working feature.

That would be awesome

It isn't really possible, since the GameCube and Wii use sampled audio. Essentially, instead of having the game's music grouped together in discrete audio tracks, each note is a sample, and the game builds the music by playing the individual notes. It saves a lot of space!
So, that means to replace a music track, you would need to add or replace the samples, and also replace the "sheet music" within the game's code. It's certainly doable if you alter the ISO, as mods have shown, but it's not like custom textures where you can replace a texture with another texture very cleanly. It would be very complicated and much, much more involved.
