The quad core would be VERY SLIGHTLY faster, like 5-10%. Dolphin does benefit from having more than 2 cores, but barely, it really has only two main threads, the other threads use very very little processing power by comparison. And that cpu would put you over budget.
As for the video card. No. Dolphin does not need that much vdram. However since it's only $5 more it's up to you.

My build includes a $26 aftermarket air cooler that is miles better than the stock pos. If you're going to spend extra money it might as well give a significant improvement.
If he kicked his budget up to the $550-575 range he could get a core i3 + P55 setup, which would be a lot better.

well yes, it seem like Dolphin or Windows like to share the work loads equally on each core. I did tried to do the "affinity" thing to force Dolphin to use only 2 cores, but it seem like it lag and stutter more that way. So letting it use all 4 cores shared is much better.
