In the controller configs, there's tabs for each controller. If the controllers you're using are detected, just select them in the dropdown menu in the upper left. If you are talking about using multiple wiimotes I'm not sure, never tried more than 1.

Yes, get 4 Wiimotes, PC, PS3, or 360 controllers.
Or get three of the above and use a keyboard/mouse for the 4th player.
